Metronomic 5-Fluorouracil Delivery Primes Skeletal Muscle for Myopathy but Does Not Cause Cachexia

چکیده

Skeletal myopathy encompasses both atrophy and dysfunction is a prominent event in cancer chemotherapy-induced cachexia. Here, we investigate the effects of chemotherapeutic agent, 5-fluorouracil (5FU), on skeletal muscle mass function, whether small-molecule therapeutic candidate, BGP-15, could be protective against chemotoxic challenge exerted by 5FU. Additionally, explore molecular signature 5FU treatment. Male Balb/c mice received metronomic tri-weekly intraperitoneal delivery (23 mg/kg), with without BGP-15 (15 6 times total over 15 day treatment period. We demonstrated that neither 5FU, nor combined affected body composition indices, or function. Adjuvant did, however, prevent 5FU-induced phosphorylation p38 MAPK p65 NF-B subunit, signalling pathways involved cell stress inflammatory signalling, respectively. This as associated mitoprotection. reduced expression key cytoskeletal proteins, desmin dystrophin, which was not prevented BGP-15. Combined, these data show does elicit physiological consequences to function but implicit priming for myopathy. has modest efficacy changes induced

Prevention of Skeletal Muscle Myopathy

Heart failure is a syndrome leading to a skeletal myopathy with muscle atrophy and shift toward fast contracting fibres. It has been shown that the major cause of atrophy is muscle waste due to skeletal muscle myonuclei apoptosis. Apoptosis is triggered by circulating cytokines and their second messengers, in particular sphingolipids. We report here several attempts to block apoptosis. Thalidom...

Skeletal muscle loss: cachexia, sarcopenia, and inactivity.

Loss of skeletal muscle mass occurs during aging (sarcopenia), disease (cachexia), or inactivity (atrophy). This article contrasts and compares the metabolic causes of loss of muscle resulting from these conditions. An understanding of the underlying causes of muscle loss is critical for the development of strategies and therapies to preserve muscle mass and function. Loss of skeletal muscle pr...

Glutamine supplementation promotes anaplerosis but not oxidative energy delivery in human skeletal muscle.

The aims of the present study were twofold: first to investigate whether TCA cycle intermediate (TCAI) pool expansion at the onset of moderate-intensity exercise in human skeletal muscle could be enhanced independently of pyruvate availability by ingestion of glutamine or ornithine alpha-ketoglutarate, and second, if it was, whether this modification of TCAI pool expansion had any effect on oxi...

Point:Counterpoint: Skeletal muscle mechanical efficiency does/does not increase with age POINT: SKELETAL MUSCLE MECHANICAL EFFICIENCY DOES INCREASE WITH AGE

We thank Dr. Ortega for the opportunity to debate the concept that aging affects exercise efficiency. From the outset we must acknowledge that, indeed, some studies report an age-related reduction task efficiency during walking and other rather complex tasks (2, 12, 20, 21). However, in contrast, our recent study of leg cycling in centenarians (28), as well as other work focused more specifical...

Prolonged treadmill training increases HSP70 in skeletal muscle but does not affect age-related functional deficits.

Skeletal muscle atrophy and weakness are major causes of frailty in the elderly. Functional deficits in muscles of old humans and rodents are associated with attenuated production of heat shock proteins (HSPs) after exercise, and transgenic overexpression of HSP70 reverses this functional decline. We hypothesized that training would increase HSP70 content of muscle in adult and old wild-type mi...
